Snap-8

Also known as: Acetyl Octapeptide-3, SNAP-8

An octapeptide extension of Argireline (Acetyl Octapeptide-3) that provides enhanced anti-wrinkle effects by inhibiting SNARE complex formation. More potent than Argireline for reducing expression lines.

Research Areas

Reduces expression lines and dynamic wrinkles
Inhibits facial muscle contractions
Improves skin smoothness and texture
Enhances skin firmness appearance
Supports collagen preservation

Side Effects

Mild skin irritation or redness
UncommonMild

May occur in individuals with sensitive skin or during initial use. Typically resolves within 1-2 weeks as skin acclimates. Reduce application frequency if irritation persists.

Dryness or tightness
UncommonMild

Some users report mild dryness, particularly if applied to already dry skin. Ensure adequate moisturization following application. Use a hydrating serum or moisturizer.

Allergic contact dermatitis
RareModerate

Rare but possible in individuals with peptide sensitivity or allergy to formulation ingredients. Perform patch test before full application. Discontinue immediately if rash, itching, or swelling occurs.

Photosensitivity
RareMild

Some peptide formulations may increase sun sensitivity. Always use SPF 30+ sunscreen during daytime use, especially when applying Snap-8.

Temporary stinging or tingling
UncommonMild

May occur immediately after application, particularly on sensitive or compromised skin. Usually subsides within minutes. Avoid application to broken or irritated skin.

Reconstitution Guide

Do not use saline or bacteriostatic saline — use only bacteriostatic water for reconstitution

Do not shake the vial vigorously; gentle swirling prevents peptide degradation

Discard immediately if the solution appears cloudy, discolored, or contains visible particles

Use within 30 days of reconstitution when stored at 2–8°C

Do not freeze the reconstituted solution; freezing may denature the peptide

Snap-8 is an octapeptide that mimics the N-terminal region of SNARE proteins, which are responsible for acetylcholine release at the neuromuscular junction. By competitively inhibiting SNARE complex formation, Snap-8 reduces acetylcholine-mediated muscle contractions in facial muscles, thereby decreasing the formation and depth of expression lines. This mechanism is similar to botulinum toxin but operates through a different biochemical pathway and is non-toxic.

SNARE Complex Inhibition

Snap-8 blocks the assembly of SNARE (Soluble N-ethylmaleimide-sensitive factor Attachment Protein Receptor) complexes, which are essential for synaptic vesicle fusion and neurotransmitter release. By preventing SNARE complex formation, the peptide reduces acetylcholine release at the neuromuscular junction.

Acetylcholine Reduction

Decreased acetylcholine availability leads to reduced muscle contraction intensity in facial muscles responsible for expression lines, particularly in the forehead, between the brows, and around the eyes.

Collagen and Elastin Support

By reducing repetitive muscle contractions, Snap-8 may indirectly support collagen and elastin preservation, as chronic muscle tension contributes to collagen breakdown and skin aging.

  • Snap-8 is an 8-amino acid synthetic peptide derived from the N-terminal region of SNARE proteins
  • It functions as a competitive inhibitor of SNARE complex assembly, reducing neurotransmitter release without systemic toxicity
  • Snap-8 is more potent than its predecessor Argireline (Acetyl Octapeptide-3) due to its extended structure
  • The peptide works topically and does not require systemic absorption for efficacy
  • Clinical studies demonstrate visible reduction in expression lines within 4-8 weeks of consistent use
